SimpleChIP® PCR引物对照

 SimpleChIP®对照PCR引物是两个正反向对照引物的混合物,用于扩增用ChIP分离出来的DNA。这些引物可以扩增阳性对照DNA序列,包括ChIP分析中抗体检测出来的目的蛋白的已知结合位点,也可以用来做阴性对照来证明抗体的敏感性。

图1.扩增--采用 SimpleChIP ® Enzymatic Chromatin IP Kit (Magnetic Beads) #9003从交联的细胞中分离DNA,然后用SimpleChIP® Human GAPDH Promoter Primers#4471进行测试。在RT-PCR检测系统中采用SYBR ® Green反应体系,梯度稀释2%的总 input DNA (20 ng, 4 ng, 0.8 ng,和0.16 ng)进行重复RT-PCR。在RT-PCR 中,PCR的扩增效率(E)和相关系数(R2)是用每一个稀释样本经40个循环的相应的阈值周期(CT)值计算出的(95°C 变性15 秒,,65°C 退火/延伸 60 秒)。

图2.特异性--从采用SimpleChIP® Human GAPDH Promoter Primers #4471的RT-PCR中得到PCR溶解曲线。数据是采用20ng总DNA两次PCR得到的。溶解曲线包括80个循环,起始温度为55度,每个循环增加0.5度。单一PCR产物的降解形成一个峰值。
