Background

蛋白泛素化作用需要E1, E2, 和E3泛素-结合酶的协同作用。泛素最先通过具有硫酯的泛素激活酶E1通过ATP-依赖形式活化。活化的泛素然后通过含有巯基的泛素载体蛋白E2转运。最后从E2的泛素转运到含有ε-氨基的赖氨酸残基靶蛋白,这由泛素连接酶E3介导(1)。

UBE2L3,通常也被称为UBCH7,是一个由四个人类相关基因组成的UBE2L1-UBE2L4泛素化结合酶家族的成员。UBE2L3是此基因家族中唯一被转录和翻译的功能蛋白,其他基因为假基因(2,3)。 作为泛素化结合酶, UBE2L3通过与多种泛素E3连接酶,如NEDD4(4)、E6AP(5)、Parkin (6)、c-Cbl (7)和 Triad1 (8,9)等相互作用参与多种底物的泛素化修饰。也有报道推测UBE2L3调解多个核激素受体(如糖皮质激素受体超家族成员、孕激素受体、雄激素受体和维甲酸受体)的转录活性(10). 另外,UBE2L3蛋白水平受到泛素相关蛋白酶体途径调解,与其参与细胞进入S期和发展有关(11)。
