Background

泛素酶(UBEs)催化蛋白质泛素化,去泛素化酶(DUB)作用相反(1,2)。共有5个亚家族的去泛素化酶被发现,包括USP, UCH, OTU, MJD和JAMM酶。USP1 属于USP家族成员,被细胞周期依赖性的转录机制和泛素——蛋白酶体系统共同调节(3)。 USP1是定位于染色质的核蛋白,特异性的与FANCD2相偶联,参与单泛素化的FANCD2的去泛素化,在DNA损伤修复和CHK稳定性调节中发挥重要作用(3,4)。USP1另一个重要的靶基因是PCNA,USP1参与单泛素化的PCNA的去泛素化,负调节PCNA介导的DNA损伤修复过程中的反式缺口合成(TLS)(5)。USP1的去泛素化活性需要与UAF1(一个含有WD40重复序列的蛋白质)相互作用并形成一个USP1/UAF1复合体(6,7)。
