Background

与泛素化修饰类似,靶蛋白Nedd8修饰通过一个包含Nedd8特异性E1活化酶和E2连接酶的酶系统介导发生(1,2)。 Ubc12就是一个Nedd8特异性的E2连接酶(3-5)。Ubc12与NEDD8形成异源二聚体,催化NEDD8 从E1转移到目标蛋白的赖氨酸残基位点(1,2)。已知的NEDD8靶蛋白属于cullin家族E3连接酶。 cullin的Nedd化修饰通过促进其与泛素E2连接酶的结合以及催化下游靶蛋白的泛素化来激活相关的泛素E3连接酶复合体(6-7)。SCFROC1、SCFβTrCP和SCFSKP2等复合体中cullins的Nedd化修饰对于一些重要的信号传递分子的调控是必需的,如IκB、NF-κB和p27 Kip(8-10),从而调节细胞周期进程、信号级联途径和正常发育过程(11)。
