Background

蛋白泛素化是重要的调节蛋白功能与命运的翻译后修饰(1)。泛素化修饰可以通过单泛素化或多泛素化形式偶联到靶蛋白,在泛素分子中有多个赖氨酸残基可以作为多泛素化链的连接位点。不同的多泛素化修饰参与靶蛋白不同的功能,包括改变蛋白功能或导致降解(2)。UBE2N/Ubc13是一个催化K63位点多泛素链形成的泛素E2连接酶(1,2)。UBE2N与MMS2或Uev1A形成异源二聚体,发挥其E2泛素连接酶功能。UBE2N/MMS2和UBE2N/Uev1A异源二聚体催化不同形式的蛋白泛素化修饰,参与信号通路的调节(3-5),包括:DNA 损伤与重组、p53和细胞周期检测点控制(6-10)、免疫受体相关信号通路(11,12)和细胞内吞过程(13)。特别是最近的研究发现,UBE2N会通过促进K63偶联的泛素化在IL-1β受体信号传递中IKK的下游级联途径等免疫相关信号通路中发挥重要作用(14)。而且, UBE2N与Triad1 E3蛋白泛素酶相互作用,在造血过程中发挥重要作用(15)。
