Background

泛素是一个在泛素——蛋白酶体途径中发挥重要作用的保守多肽。泛素能够通过泛素转移过程被共价修饰于多个细胞蛋白,导致蛋白通过26S蛋白酶体降解。三个泛素化酶参与泛素偶联过程:泛素首先与活化酶E1(UBE1或E1)形成硫酯键复合体而被激活,然后被转移到泛素载体蛋白E2,然后由E2转移到E3并连接到靶蛋白赖氨酸的epsilon位氨基上(1-3),不同E2和E3的组合决定底物特异性(4)。UBE1有两个亚型:UBE1a是一个117kDa的核蛋白,UBE1b是一个110kDa的核质蛋白(5).
