Background

泛素酶(UBEs)催化蛋白质泛素化,去泛素化酶(DUB)的作用相反。共有5个亚家族的去泛素化酶被发现,包括USP, UCH, OTU, MJD和JAMM酶(1,2)。USP18 (也称UBP43)是研究最为深入的参与催化ISG15从靶蛋白移除的去泛素化酶的,并受到干扰素调节(3)。USP18介导ISG15从靶蛋白移除对于维持参与正常发育过程和脑功能的ISG15修饰蛋白的平衡至关重要(4,5)。在IFN或LPS诱导下 (6),USP18结合到干扰素受体亚基IFNAR2并抑制JAK-STAT信号通路传递(7)。USP18对干扰素信号的调节,抑制干扰素引起的细胞凋亡,并且这种调节不依赖于USP18肽酶活性(8)。重组干扰素在治疗中使用会导致干扰素信号通路响应下降,因此将干扰素治疗与表达USP18相结合可能获得更好的疗效(9)。
