Background

CYLD是一种由一种肿瘤抑制基因编码的细胞质去泛素化酶,在圆柱瘤患者中该基因发生改变,该疾病是一种由基因变化引起的皮肤附件的良性肿瘤(1,2)。功能性的CYLD去泛素化酶,通过去除几种NF-κB通路蛋白的泛素链,下调NF-κB信号转导,进而调节炎症和细胞增殖(3,4)。CYLD是邻近Wnt/β-catenin信号传导途径的一种负向调节因子,并是自然杀伤T细胞发育中的一种重要的调节因子(5,6)。转录因子Snail能够抑制CYLD的表达,从而导致黑色素瘤细胞增殖(7)。

CYLD can be phosphorylated at Ser418 by the noncanonical IKK family member IKKε. Phosphorylation at Ser418 decreases CYLD deubiquitinase activity and is important for IKKε-driven transformation (8).
