Background

蛋白泛素化需要泛素结合酶E1、E2和E3协调一致的作用泛素首先与活化酶E1形成硫酯键复合体而被激活,然后被转移到泛素运载酶E2,然后由E2转移到E3并连接到靶蛋白赖氨酸的epsilon位氨基上(1)。UbcH5C是泛在表达的Ubc家族E2泛素结合酶(此家族还包含UbcH5A和UbcH5B)(2)。有证据提示UbcH5C通过催化关键蛋白质分子(包括p53、PCNA、IκB激酶蛋白NEMO、凋亡抑制分子BRUCE等)的泛素化修饰而在多个信号通路调节中发挥重要作用(3-6)。基因表达分析显示UbcH5C在睑板腺细胞癌和嗜酸性甲状腺腺瘤表达升高 (7,8),而通过RNAi筛选试验发现急性早幼粒细胞中Ubc5HC存在差异化表达(9)。这些结果提示Ubc5HC在细胞周期调控和肿瘤发生中发挥作用。
