CDK-activating kinase (CAK) is a complex of CDK7 and cyclin H. The complex is involved in cell cycle regulation by phosphorylating an activating residue in the T-loop domain of cdks (1). Regulation of CAK activity is mediated by T-loop phosphorylation and by association with MAT1, both of which enhance its kinase activity toward the CTD of RNA polymerase II (2,3) and other substrates such as p53 (4). CAK is an essential component of the transcription complex TFIIH and may interact directly with TFIIH helicases (5). CDK激活激酶(CAK)是CDK7和周期蛋白H的复合体。该复合体通过磷酸化cdks的T-回路结构域的激活残基参与细胞周期的调节(1)。CAK活性的调节经由T-回路的磷酸化和MAT1的缔合来介导,这两个过程都能够增强CAK对于RNA 聚合酶II(2,3)的CTD以及其它底物如p53(4)的激酶活性。CAK是转录复合体TFIIH必不可少的组成部分,并可以直接和TFIIH解旋酶相互作用(5)。
